This study was a prospective, randomized, active-controlled, double-masked, phase 3 trial conducted from August 6, 2004, to October 6, 2005, at 47 sites. Subjects with a clinical diagnosis of bacterial conjunctivitis were randomly assigned to receive either 1% azithromycin in Dura Site (Aza Site; In Site Vision, Alameda, CA) ( = 378). Both groups received masked medication four times daily for 5 days, but participants received an active dose of 1% azithromycin in Dura Site only twice a day on days 1 and 2 and daily on days 3 to 5. Conjunctival cultures were taken, and ocular signs and symptoms were evaluated at baseline and at two follow-up visits. A total of 743 patients were randomized, and 710 (96%) completed the trial. The most frequently observed ocular adverse events in the azithromycin group were eye irritation (1.9%), conjunctival hyperemia (1.1%), and worsening bacterial conjunctivitis (1.1%). These rates compared favorably with those obtained with tobramycin. Rates of microbial eradication (an efficacy parameter) and bacterial infection recurrence (a safety parameter) were the same in both groups. Known hypersensitivity to azithromycin or erythromycin Serious allergic reactions, including angioedema, anaphylaxis, and dermatologic reactions including Stevens Johnson Syndrome and toxic epidermal necrolysis have been reported; although rare, fatalities have been reported Discontinue use if superinfection occurs May result in overgrowth of nonsusceptible organisms including fungi Avoid contact lens wear during therapy of S/S of bacterial conjunctivitis Do not administer systemically; inject SC; or apply directly into anterior eye chamber The above information is provided for general informational and educational purposes only. Azithromycin helps to kill the bacteria which are causing the infection. Eye infections are a common cause of conjunctivitis. In conjunctivitis, your eye becomes inflamed, feels gritty, and may water more than usual. The white of your eye may look red, and your eyelids can become swollen and stuck together with a discharge when you wake up in the morning. Patients and methods This was a multicentre, international, randomised, investigator-masked study in 286 children with purulent discharge and bulbar conjunctival injection. Patients received either azithromycin 1.5% eye drops (twice daily for 3 days) or tobramycin 0.3% eye drops (every 2 h for 2 days, then four times daily for 5 days). Clinical signs were evaluated on day (D) 0, 3 and 7, and cultures on D0 and D7. The primary variable was the clinical cure (absence of bulbar conjunctival injection and discharge) on D3 in the worse eye for patients with positive cultures on D0. Results 286 patients (mean age 3.2 years; range 1 day–17 years) were included; 203 had positive cultures on D0. Azithromycin was superior to tobramycin in clinical cure rate on D3 (47.1% vs 28.7%, p=0.013) and was non-inferior to tobramycin on D7 (89.2% vs 78.2%, respectively). Azithromycin treatment eradicated causative pathogens, including resistant species, with a similar resolution rate to tobramycin (89.8% vs 87.2%, respectively).

β adrenergic receptor antagonists (also called beta-blockers or β-blockers) were initially developed in the 1960s, for the treatment of angina pectoris but are now also used for hypertension, congestive heart failure and certain arrhythmias. In the 1950s, dichloroisoproterenol (DCI) was discovered to be a β-antagonist that blocked the effects of sympathomimetic amines on bronchodilation, uterine relaxation and heart stimulation. Although DCI had no clinical utility, a change in the compound did provide a clinical candidate, pronethalol, which was introduced in 1962. The β-blockers are an immensely important class of drugs due to their high prevalence of use. The discovery of β-blockers reaches back to more than 100 years ago, when early investigators came up with the idea that catecholamines were binding selectively to receptor-like structures and that this was the cause of their pharmacological actions. Ahlquist published a seminal paper concluding his findings, that there were two distinct receptors for catecholamine drugs, and they caused different responses in the heart muscle. These findings were soon to be a foundation for further research into drug development. In the early 1960s, James Black, a Scottish pharmacologist, and associates of his at the Imperial Chemical Industries (ICI) in Great Britain were working on a series of β-adrenergic blocking compounds, pronethalol and propranolol. Black focused on developing a drug that would relieve the pain of angina pectoris, which results from oxygen deprivation in the heart. His plan was to create a drug that would decrease the heart’s requirement for oxygen. What are the warnings for taking Metoprolol Lopressor? Using condoms during sexual intercourse significantly decreases the likelihood that men infected with herpes simplex virus type 2 (HSV-2) will transmit the infection to their female partners, according to the first study to examine the effectiveness of condoms in preventing this infection. Women are almost six times as likely as men to acquire HSV-2. Increased frequency of sexual intercourse, younger age and having a partner who is infected with both herpes simplex virus type 1 (HSV-1) and HSV-2 increase the likelihood of acquiring HSV-2. Although using condoms more than 25% of the time offers women a high degree of protection against acquiring HSV-2, men do not receive the same benefits. To assess whether using condoms reduces the transmission of HSV-2, researchers analyzed behavioral and demographic data from participants in two multisite HSV vaccine trials conducted in the mid-1990s. The study included adults who, at enrollment, tested negative for both HSV-2 and HIV ("susceptible partners"), and had been involved in a monogamous relationship for at least six months with an individual infected with HSV-2 ("source partners"). Susceptible partners were interviewed during an initial screening, where they were instructed to keep a diary of their sexual activity for the duration of the study.
