    Paramagnetic metal centers such as FeIII found within ferriprotoporphyrin IX heme FPIX exert through space effects on the relaxation rate of nearby proton spins that depend critically on the metal−proton distance. We have measured these effects for all protons of several antimalarial drugs that bind to FPIX by systematically varying the drugheme molar ratio in high field NMR experiments. Absorbance spectroscopy provides evidence that chloroquine is capable of extracting haem bound to Pfhrp-2. This was supported by the Kd value, of 37 nM, observed for the haem-chloroquine complex. The native PAGE studies reveal that the formation of the haem-Pfhrp-2 complex is disrupted by chloroquine. In the presence of free heme, chloroquine and quinidine associate with the heme polymer. We have proposed that this association of the quinoline-heme complex with polymer caps the growing heme polymer, preventing further sequestration of additional heme that then accumulates to levels that kill the parasite.

    Chloroquine possesses definite antirheumatic properties. Chloroquine has immuno-modulatory effects, suppressing the production/release of tumour necrosis factor and interleukin 6. Chloroquine inhibits trophozoite hemoglobin degradation through increasing vacuolar p H and inhibiting the activity of vacuolar phospholipase, vacuolar proteases, and heme polymerase.

    Chloroquine is a chemotherapeutic agent for the clinical treatment of malaria. Attenuated total reflectance infrared spectroscopic data clearly show that heme in chloroquine-FP complex is not μ-oxo dimeric indicating that the hypothesis that chloroquine binds to FP μ-oxo. Chloroquine is thought to exert its antimalarial effect by preventing the polymerization of toxic heme released during proteolysis of hemoglobin in the Plasmodium digestive vacuole. The mechanism of this blockade has not been established. We incubated cultured parasites with subinhibitory doses of 3Hchloroquine and 3H quinidine. These 3Hquinoline compounds became associated with hemozoin.
