No effective prophylactic or post-exposure therapy is currently available. We report, however, that chloroquine has strong antiviral effects on SARS-Co V infection of primate cells.

The combination of chloroquine with zinc enhanced chloroquine’s cytotoxicity and induced apoptosis in A2780 cells. Thus chloroquine is a zinc ionophore, a property that may contribute to chloroquine’s anticancer activity. Chloroquine inhibits the intracellular multiplication of Legionella pneumophila by limiting the availability of iron. Chloroquine CQ is a lysosomotropic agent with an extensive range of biological effects 1. Historically known for its anti-malarial activity, chloroquine is a widely used biological research tool for studying autophagy inhibition. Research studies demonstrate that chloroquine accumulates in acidic lysosomes and increases the lysosomal pH. I have tried daily change with medium with new chloroquine for thje autophagy studies and keep the same culture medium and chloroquine for 24-72 hours so far I have not seen difference in the results.

In addition to the well-known functions of chloroquine such as elevations of endosomal p H, the drug appears to interfere with terminal glycosylation of the cellular receptor, angiotensin-converting enzyme 2. These inhibitory effects are observed when the cells are treated with the drug either before or after exposure to the virus, suggesting both prophylactic and therapeutic advantage.

  7. Chloroquine is an inhibitor of the lysosomal degradation of the DNA which is taken up by the cells, so as leelee said, transfection should have been successful, albeit at a slightly lower level than if you would have added the chloroquine. In my normal transfections, I never add chloroquine and get high efficiencies nevertheless.

Inhibition of Zika virus ZIKV infection by chloroquine in Vero cells. A Vero cells were infected with ZIKV MR766 at a multiplicity of infection MOI of 2, treated with chloroquine for 5 days, and were then stained for the viral envelope protein and. Jan 27, 2010 Chloroquine CQ, traditionally used as an antimalarial medication, has emerged as a prospective adjunct in tumor therapy. CQ has been found to strongly potentiate the inhibitory effect of radiation on cancer cell proliferation, presumably via negative regulation of DNA repair processes. Nov 29, 2016 Chloroquine reduced 45% and 50% of the number of ZIKV MR766-infected hBMECs at 25 and 50 μM, respectively Figure 4 A,D. These concentrations are non-cytotoxic Figure 4 B, and protected approximately 80% of hBMECs from ZIKV infection as demonstrated by the increase in cell viability Figure 4 C.

And as far as the vision issue goes, toxicity causing vision loss is very rare if you are on the right dose for your height and if you get regular eye exams. And those who do get toxicity have usually been on the drug for years and years.
